Zusammenfassung

Pine pitch canker (PPC) is a serious disease affecting Pinus spp. and Pseudotsuga menziesii worldwide. Several factors have contributed to the spread of the disease to all these continents, the most important being the international trade in plant reproductive material. Despite its importance, no effective measures are available to eradicate or control the disease either in nurseries or in the field. In this thesis, a compilation of current information on the pathways of spread of the pathogen was carried out, proposing preventive mechanisms to avoid its introduction into disease-free areas. We propose further research on the possible role of mycoviruses as biological control agents, exploring new methods of transfection with F. circinatum mycoviruses. We used transcriptomics technologies that, in addition to allowing us to study the effect of a mycovirus on its host fungus at the molecular level, provided useful information on genes, transcripts, and their functions associated with plant-pathogen interactions. Using RNA-Seq and susceptible and resistant species to F. circinatum, we identified genes and metabolic pathways associated with disease resistance traits and presented a comprehensive mapping of lncRNAs during pathogen infection. Our findings on the regulatory mechanisms involved in host defence will allow the optimisation of breeding programmes for the commercialisation of PPC-resistant pines.
